Abstract

Identifikacija parazita na temelju morfoloških obilježja i dalje se smatra zlatnim standardom u klasičnoj kliničkoj dijagnostici. Međutim, moguće su pogreške u identifikaciji zbog male količine dostupnog uzorka, uzorka koji ne obuhvaća parazita u svim životnim stadijima te činjenice da su određeni paraziti morfološki vrlo slični. Stoga je cilj našega rada bio molekularnim metodama provjeriti pouzdanost morfološke identifikacije crijevnih parazita prikupljenih iz 23 jedinke čaglja (Canis aureus)... More iz Hrvatske. Utvrdili smo da su uzorci dvije vrste oblića pravilno identificirani (Toxocara canis, Toxascaris leonina), dok kod vrste Alaria alata nismo uspjeli provesti molekularnu identifikaciju. Molekularna dijagnostika je pokazala da uzorak morfološki identificiran kao oblić Uncinaria stenocephala zapravo pripada vrsti Toxocara canis, dok uzorak morfološki identificiran kao mala pasja trakavica Echinoccocus granulosus pripada vrsti Echinoccocus multilocularis. Dobiveni rezultati osobito su značajni sa stajališta javnog zdravstva, s obzirom da bolesti koje navedeni paraziti uzrokuju zahtijevaju različiti pristup u kontroli. Stoga u slučajevima važnim za javno zdravstvo ili kod moguće pojave novih vrsta preporučujemo provjeru pouzdanosti rezultata upotrebom molekularnih metoda. Abstract (english)

Identification of parasites based on their morphology is still considered as the gold standard in classic clinical diagnosis. However, there is a possibility for misidentification because of low quality and quantity of sample, sample which does not include all life stages of parasite or the fact that certain parasites are very similar in their morphological characteristics. So the goal of our research was to test the reliability of morphological identification of intestinal parasites found in... More 23 jackals (Canis aureus) from Croatia. We confirmed that Toxocara canis and Toxascaris leonina samples were identified correctly, while quality of results of our molecular identification were not good enough to confirm the identification of Alaria alata. Molecular method showed that the nematode morphologically identified as Uncinaria stenocephala actually is Toxocara canis, while Echinoccocus multilocularis was wrongly identified as Echinoccocus granulosus. Considering the differences in pathogenesis and treatment of the diseases caused by these parasites, our results are very important for public health. So we recommend to confirm morphological identification with molecular methods when diagnosing important parasitic zoonosis and describing new species. Our results are also significant because this is the first identification of Echinoccocus multilocularis in jackals on Croatian territory. Less
