Abstract

Tijekom sedam dana diferencijacije in vitro stanice su se u potpunosti diferencirale od
progenitora oligodendrocita do potpuno zrelih, mijelinirajućih oligodendrocita. Prvog dana
diferencijacije 99% stanica je olig2, sox2 i nestin pozitivno. Već prvog dana diferencijacije
88±2% stanica je o4 i o1 pozitivno, dok je svega 10% stanica gfap pozitivno. Trećeg dana
diferencijacije udio olig2 pozitivnih stanica smanjuje se na 86±3% stanica dok se udio o4, o1 i
gfap pozitivnih stanica zadržava na istim vrijednostima kao i prvog dana diferencijacije. Petog
dana diferencijacije, stanice su potpuno diferencirani oligodendrociti, a udio olig2 pozitivnih
stanica iznosi svega 11±2% stanica, dok se udio o4, o1 i gfap pozitivnih stanica zadržava gotovo
na istim vrijednostima kao i u ranijim vremenskim točkama. U ovoj vremenskoj točki 10±2%
stanica je mbp pozitivno dok je 79±2% stanica plp pozitivno, pri čemu svega 12%
mijelinirajućih oligodendrocita je pozitivno na oba biljega. Sedmog dana diferencijacije udio
o4 i o1 pozitivnih stanica i dalje iznosi 82±3% stanica dok je udio gfap pozitivnih stanica
neznatno porastao i iznosi 11±3% stanica. Udio mbp pozitivnih stanica porastao je na 13±2%
stanica dok je udio plp pozitivnih stanica porastao na 84±3% stanica

Abstract (english)

During the seven days of in vitro differentiation, cells were completely differentiate
from the oligodendrocyte progenitors to the mature, myelinating oligodendrocyte. On the first
day of differentiation 99% cells were olig2, sox2 and nestin positive. Moreover, 88±2% cells
were o4 and o1 positive, while the only 10% cells were gfap positive. On the third day of
differentiation, number of the olig2 positive cells was reduced to 86±3% cells while the o4, o1
and gfap positive cells hold at the same values as on the first day of differentiation. On the fifth
day of differentiation, cells were completely differentiated oligodendrocytes and number of
olig2 positive cells was reduced to 11±2% of cells, while the proportion of o4, o1 and gfap
positive cells holds almost at the same values as in earlier time points. At this time point, 10±2%
cells were mbp positive while the 79±2% cells were plp positive. Taken all together only 12%
of myelinating oligodendrocyte were positive on both markers. On the seventh day of
differentiation, number of o4 and o1 positive cells were 82±3% of cells, while the number of
gfap positive cells was slightly increased to 11±3% positive cells. The percentage of mbp
positive cells were increased to 13±2% cells, while the percentage of plp positive cells were
increased to 84±3% cells.
