Abstract

Cilj svake uspješne proizvodnje peradi je zadržati zdravlje jata. No upravo su mlade jedinke s najizraženijim potencijalom rasta najpodložnije zarazi patogenima. Probavni sustav je zbog svoje funkcije stalno izložen raznim mikroorganizmima od kojih su neki i poželjni dok oni drugi svojim štetnim djelovanjem izravno kočr rast i razvoj same jedinke. U ovom istraživanju analiziran je utjecaj astrovirusne infekcije na serumske proteine purića s ciljem pronalaženja potencijalnih markera kojima bi se moguće patološke promjene detektirale već prije početka bolesti. Za ovo istraživanje korišteni su serumi 15 purića te uzorci slezene i tankog crijeva 5 purića, linija Hybrid, dobi 5 I 13 dana. Prethodnom dijagnostikom su u uzorcima skezene i tankog crijeva, metagenomikom primjenom sekvenciranja nove generacije, dokazani astrovirusi u jata dobi 13 dana te odsustvo virusa u drugom jatu, koje je korišteno kao negativna kontrola prilikom kasnije analize proteome seruma. Za identifikaciju i relativnu kvantifikaciju proteina seruma korišten je SEQUEST algoritam unutar računalnog programa Proteome Discoverer. Kako bi se protein točnije identificiralo i kako bi se definirala njihova uloga u organizmu koristili smo programe STRINGdb v11.0, KEGG Pathways, REACTOME Pathways. Analiza proteome pokazala je jasnu razliku proteomskih podataka jata pozitivnog na astrovirus u odnosu na negativno jato. Identificirano je 68 jedinstvenih proteina, od kojih je 46 pojačano eksprimirano, a 22 podeksprimirano, te se moguće mogu koristiti kao dijagnostički marker za dokaz i praćenje zaraze astrovirusom ili drugim crijevnim ili sistemskim virusima.

Abstract (english)

The goal of every successful poultry industry is maintaining the health of the flock. But, young birds with the most prominent growth potential are the category that is the most prone to infection. Gastrointestinal tract is, because of its function, constantly exposed to various microorganisms, some of which are desirable, while others are harmful to the growth and health of an individual. In this paper we conducted research on the impact of astroviral infection to the serum proteins of turkey poults, with the goal of finding potential markers for the detection of pathological changes before the clinical onset of the disease. For this research we used serums of the 15 poults and spleen and small intestine samples of 5 poults, of Hybrid genetics, of 5 and 13 days of age. Previous diagnostics on the spleen and small intestine samplesm with metagenomics using next generation sequencing, showed the presence of astroviruses in 13 days old flock, while younger flock was negative for all viruses and it was used as a negative control for later serum proteome analyses. Algorithm from the computer programe “Proteome discoverer” was used for identification and relative quantification of serum proteins. To identify proteins and their roll in organisms we used STRINGdb v11.0, KEGG Pathways, REACTOME Pathways. Proteome analysis showed a significant difference between positive and negative flock. Among 171 serum proteins with significant abundance changes between healthy and infected turkeys, 68 of them were unique proteins, from which 46 of them were overexpressed, and 22 under-expressed. In conclusion, 68 proteins could be possible markers for diagnostics and follow up of astrovirus or other enterovirus or systemic virus infection.
